1)

Why did Yaakov command the second and third messengers to tell Esav, even if he does not ask?

1.

Malbim: If Esav did not veer to go to Se'ir, but is still heading towards Yaakov, they should make it appear that they and Yaakov are going to greet him, i.e. he feels only love for his brother.

2)

Why does it say, "also all who go"? This should include more than just a fourth and fifth (group), because the word "Kol" means at least three. But there were only 5 types of animals!

1.

Ha'amek Davar: the word "all" refers to all the messengers. He sent several slaves with each herd, according to the need.

3)

Why did he say, "ka'Davar ha'Zeh"?

1.

Ha'amek Davar: They cannot say exactly like the first group, for they are to say even if not in response to a question. Even though Esav already knows from the first messengers, Yaakov wants him to hear repeatedly, "to your servant Yaakov," "to my master Esav."
